Only by its active site . Let's check. We know that the Emil Fischer model which says that active site is rigid which tells us the specificity. Also Daniel model says that active site that enzyme is flexible which is important for the specificity of an enzyme. Understand...???

active sites because if the shape of substrate matches the active site then the reaction can proceed
